The Pro Roller Bag 70 from Manfrotto holds two DSLRs with attached lenses such as a 24-70mm and 70-200mm, six additional lenses, and two flashes between touch-fastened red rigid dividers and gray padded flexi-dividers inside the main compartment. The Roller Bag also accommodates a 17" laptop behind a padded divider on the inside of the front flap and an iPad or similar sized tablet inside a zippered pocket on top of the laptop divider. question
Can you recommend a Manfrotto top case or shoulder bag that would slide in between the handles and rest on the bag?
The Manfrotto Pro Shoulder Bag 10 B&H # MAPSB10 would have a trolley strap. Please see the image for reference.

question
Is the laptop compartment padded?
It's likely too late to help but yes the laptop area is padded (albeit lightly). I would say it is better protected from the inside rather than a sharp hit to the outside front... On a side note I use this case from air travel and it fits in the larger over head bins but not all of them! Some smaller planes are tough. Even the larger ones require a good slam of the door...

question
Does The Manfrotto Pro Roller Bag 70 have locks or can travel locks be used?
It does not have locks, but the sturdy zippers for the main compartment have a dedicated spot for a TSA style (or similar) lock.

question
I'm on the fence between this and the Tamrac 5592 Big Roller 2X? How do they compare in terms of build quality?
Bought this in June 2014. Not sure about the Tamrac, but the MAnfrotto case has been wonderful. Lots of space, we can fit all out gear in it and it's been strong quality build.

question
Will this bag fit into standard airplane over hear compartment?
Currently the FAA rules state that the maximum size carry-on bag for most airlines is 45 linear inches (the total of the height, width, and depth of the bag). Anything larger should be checked. The Manfrotto Pro Roller Bag 70 has a total of 46 linear inches. In situations like this my best advice is that you contact the airline you will be traveling on, and they can advise what their current size restriction may be, and if any exceptions can be made or not.
