The 12" Jerkstopper Zip from TetherTools provides your cables with an anchor point allowing slack in the cord or cable to prevent stress, dislodging, and damage. The steps used to apply it are just reversed. There are 2 components that slid together, they can also be adjusted for tighter or looser applications.

Will this work to hold a remote on a Canon 5D Mark2?

We purchased this item (actually 3 of them) for video camera use, and it's great. Basically it is designed to remove cable tension. With that in mind, for most small to medium size cables of any nature it should work.

Can the very thin cord on the Tether Tools JerkStopper be replaced by the user with a stronger, more heavy duty cord?

Bottom line, no. The cord can not be removed from the one side of the clip. But I agree that it is very strong and I have no concerns about it's function.
