What's the major difference between this and the GFX 50R? Is it worth $1000 price difference?

The 50R is smaller and lighter. The 50R has a smaller grip. On the 50R the viewfinder is on the left side of the camera (rangefinder style), and the 50S it is located at the center (SLR design). The EVF on the 50S can be removed, or oriented at different angles with the optional EVF-TL1 tilt adapter. The rear monitor on the GFX 50S can be tilted in three ways just like the X-T2, X-T3 and X-H1 cameras. In addition to the traditional up and down movements, you can tilt it to the right by approximately 60°. They both have wifi but the 50R has bluetooth. The GFX 50S can be used with an optional battery grip (VG-GFX1) to double the battery life of the camera. No vertical grip has been developed for the 50R.

I agree with the others: It May be possible to find an adapter that would allow you to physically mount X lenses on the GFX bodies, but those lenses are not designed to project an image across the entire, larger sensor. The more common situis to go the other way by putting legacy medium format lenses on the GFX cameras via adapters.

My GFX 50S was just delivered; I'm setting up ...

My GFX 50S was just delivered; I'm setting up custom functions. No where in the manual does it specify, but do WB, Film Simulation and DR not show in the LCD and EVF when they are set to auto or standard? I assume this is the case as they show when they are on a different setting. Seems odd, perhaps I'll get used to it. Thanks.

It is common for cameras to block out certain manual overrides when a camera is set to auto mode. Fuji only allows you access to the WB, film simulation and DR modes in the more manual settings.
