They plug into different locations. A wireless flash trigger would be mounted on the hot shoe, while a remote control would be plugged into the port on the left. Once the shutter is enabled, a flash would fire.

With the GFX 100 its required to update the lens firmware. So If I update my current GFX Lens used with my 50R firmware, will the lens still work with my GFX 50R. Are the lenses interchangeable with the firmware update ?

Fuji released new firmware for all the lenses back in June 2019, which added phase detection AF compatibility, so the lenses will focus faster with GFX 100 than they do with 50S or 50R. After the firmware upgrade, the lenses will still work with older GFX camera bodies.
