Will the record start/stop button function via the canon EF mount if the hand unit is powered by D-tap and does the Canon C100 MK2 EF mount communicate with this lens?
Unfortunately it will not. For that you would need a camera with a 12 Pin Control Cable, such as the Canon C700.

If Auto focus is available when using this lense on C300 Mark III, does it breathe a lot?
Yes, autofocus is available! According to the Canon C300 mkIII instruction manual the camera supports all focus modes with this lens, including continuous AF, face AF, and tracking. The Canon C100 and C300 mkII and original C300 with DPAF upgrade also support autofocus with this lens thanks to a firmware update released in December 2016. Note that the lens firmware may also need to be upgraded to support autofocus if you have older firmware.
The lens is parfocal, and minimizes breathing.
