Can this test cables with different connectors on each end? For example, XLR on one end and 1/4 TRS on the other end.
Yes it can .For stereo to stereo connectors there is no problem eg XLR 3 PIN TO STEREO 1/4 JACK. When one connector is wired in stereo and the second mono it will show you clearly the cables that are joined together for the mono connector.

Hoping to verify that the jack on the side is for accepting 1/4 (6.35 mm) . It doesn't actually say it in the specs.
I can confirm the Kopul CBT-8 8-in-1 Cable Tester can accept 1/4" TRS/TS connections to test continuity.
