1) Is this tank meant to be used together with the Jobo processing machine? Can it be used for manual processing? i.e. let the tank stand and stir at regular intervals.
2) Is the 640ml meant when the tank is used with the Jobo processing machine? If I use normal stand development, will I need to use more amount of chemicals?
The Jobo 2550 Multi Tank 5 (640mL) can be used either with the Jobo processing machine, or for manual long, stand, or semi-stand processing. 640mL is the minimum amount of chemistry required for rotary processing. For stand processing you will need more chemical.

Is this compatible with the uniformed 352?
I contacted the manufacturer concerning your inquiry. According to Jobo, in theroy, the Jobo 2550 Multi Tank 5 (640mL) may be used on the Unicolor Uniroller Model 352 Professional Photography Film Drum Roller. However, they do caution that as these two items are unrelated systems, results may vary.

How is this a 640mL tank, when the Paterson Multi-reel 3 is significantly smaller, and takes 1L of chem?
You will use 640ml of chemical when the tank is used on a rotary processor where the tank is turned on its side during processing. If you use the tank in inversion processing, stand or semi-stand developing for instance, then it will take a lot more chemical to fill it because inversion requires the tank to be upright.

If this is used by hand, without the column is it still light tight when adding chemistry?
Unfortunately, I am not sure, as this is not the normal method for using this tank without the center column. The best advice I may offer would be if you can see the opening, then the light will probably enter the tank. To be on the safe side, I would not use it in this way. Otherwise, I would recommend first practice/test with a blank roll or spare roll of junk film that has not been exposed and which you do not mind using to see if the film appears to be exposed to light during the normal processing process while not using the center column.
