Would this work on a platypod. I'm concerned that the tightening knob would not allow it to sit flat or be hard to turn?
Unfortunately, this would not work with the Platypod. In our testing the tightening knob does not allow the head to sit flat and cannot be tightened fully.

What are the functional differences between this and the Kirk TQR-1S?
The Kirk TQR-3S Tripod Head Quick Disconnect Small System with TQR-MPS Plate is for smaller tripods. It includes an assembly that measures ø: 2.0 x W: 2.8. The Kirk TQR-1S is made for wider tripods. It includes an assembly that measures ø: 2.9 x W: 4.5. I recommend purchasing a system that is closer to the size of your tripod base.
