The USB port on the Vello Battery Grip for Nikon Z6 II and Z7 II Mirrorless Camera will only charge the batteries if the camera is turned off.

will this work with the first gen ftz adapter? ... is there clearance for the extruding tripod mount?

Just purchased this battery grip. I have both the FTZ and FTZ II adapters. There is more than enough room for my pudgy fingers to grip between the base of the FTZ and the grip, so no issues there. You will definitely not be able to leave a baseplate, or any other accessory on the FTZ and use the grip (not that you would ever have any reason to.) So I believe the short answer is YES they are compatible.
