The lock mechanish of the rolltop is really robust and works smoothly, but there is no way of putting any lock to it.

Will this hold a R5 w/70-200 attached, 100-500 and a 2?4-70

Although Thinktank shows it can carry one 70-200 with a pro body like R5, but in real-world situations, the side access and the rear hatch of camera compartment have very narrow openings for the kind of usage ThinkTank markets it to. I experienced difficulties with Fujifilm X-h2S, taking it in and out of the bag with Sigma 17-40 1.8 attached.
