There are 2 holders in the image. One for 4x5 inches and the other for 9x12 cm. The reel is adjustable and can be used for both these sizes. These holders are to secure the film in place once it is loaded.

In case of use with cog lid is it necessary to use the core tube?
Absolutely, with the cog its on the outside of the tank. The core tube in necessary to maintain light from fogging the film. Needed every time regardless of the process method.

How many reels do you need for the tank?
The answer would depend on which Jobo developing tank you own. The Jobo Multi Tank 2 (270 mL), B&H # JOMT2, will accommodate one (1) Jobo 2509N Sheet Film Reel, while the Jobo 2550 Multi Tank 5 (640mL), B&H # JOMT5, would accommodate two (2) Jobo 2509N Sheet Film Reels.
