Wanting to isolate a balanced run so can I input balanced, route unbalanced output back into unbalanced input and get balanced output?

You may take a balanced line into ch 1 bal line in, out of line 1 Unbal line out into line in 2 and then back out line 2 bal out, so yes.Im not sure how it will sound tho, as thats a lot of conversions and impedance changes- better to use an isolation transformer, or buy an in line isolation transformer/barrel instead.

No, the ART CLEANBoxPro Bi-Directional Level Matching Converter is not a preamp, and will not properly amplify a mic signal coming from a 3.5mm cable.

The ART Cleanbox Pro is neither ideal, nor made for that use. You would want to use something more like B&H # SES43DBMZ2XJ or B&H # KOCMX2V2 for that application.

Will this work with the output of a Vocaster 2 (1/4") to Mic input of a ATEM Mini Pro (3.5")? If so what connectors do I need?

The ART CLEANBoxPro does not convert Line level to Mic level, so it would not be suitable for connecting the monitor outputs of an interface to the mic input of a switcher. However, its 3.5mm output could be used in conjunction with an attenuator cable such as BH #WHH4NL3.5 for this purpose.

No, there are two separate one way paths. Bal to unbal and unbal to bal.
