In order to file transfer ProRes over USB 3.0 to PC and eventually NAS, will I need to install MacDrive software on my PC?

I use this drive in the AJA Ki-Pro to record an hour-long church service, then connect via FireWire to my MacBook Pro and export the file as an .mp4 using FCP. The drive shows up in Finder just like any other external HDD, with no additional software needed.

Do I have to have a MAC to read the AJA drive?

It depends on what filesystem you set up on the drive. Historically KiPros have used only Apple's HFS+ filesystem, which is native to Macs and not native to other OSes. Those OSes, Windows and linux, can both be extended to recognize HFS+ through free or purchased software.I see that Aja has starting offering KiPros that use either the HFS+ or ExFAT filesystems. ExFAT is native to both Windows and MacOSX, and can be added to linux.So, in the end it depends on what your KiPro can do and how you use it, and perhaps how you adapt your computer OS.
