With that lens, the effect of this filter occurs behind the main glass elements instead of in front of them.

This polarizer is just for the 400mm fixed lens. The lens has a slot on the top where the filter drops in.

Do you have 5 stop and a 10 stop ND filters that will screw in to these drop in filter holders?

The Nikon AF-S NIKKOR 400mm f/2.8E FL ED VR lens came with a drop in filter holder and a clear filter. You can remove the clear filter from your original holder and use any 40,5mm filter in the holder instead such as this 5 stop ND: https://bhpho.to/4al5eNO

Is the filter adjustment ring water resistant? Seems like a place that could potentially let in water into the lens during rain.

I am not sure but its a direct replacement for the filter that comes with the lens. I use a Thinktank cover any time Im shooting where the lens could get wet.

Nikon 400mm lens come with a drop-in filter. If you want a polarizing filter, the 40.5mm drop-in filter only works in the Nikon AFS 400mm f2.8E FL ED VR. This is the current model. The older AFS 400mm f2.9 requires the 52mm Circular Polarizer (C-PL1L).
