Yes, this will. First, remove the Manfrotto 500 fluid head to reveal the 3/8"-16 screw from the XPRO monopod, then mount the 3-way pan head onto it.

While we not are familiar with the "500 monopod" (did you mean the Manfrotto XPRO monopod with the MA500 fluid head?), this will work with any monopod with a 3/8"-16 mounting thread, of which most tripods are.

I would like to use a pan and tilt head with a spotting scope, so I only need two axes of movement. Can the middle part of this head be removed and replaced with the top part (i.e., remove the part with the lower handle), to make a 2-way head?

No, the pieces are designed to go together the way that they are, I see no possibility of removing the middle piece and reassembling it differently. I think that you need to look for a 2 axis head. There are hundreds of heads out there. This is a nice head and you can independently lock each axis, so it should work for you as-if if you just leave one axis tightened up.
