Can I fit a Canon 5D3 with 24-105 lens attached, and a 50 mm lens in the insert?

I looked up all the sizes of the equipment you mentioned. The photo compartment is not a perfect cube because of manufacture tolerances. I looked at the scenario with the zoom lens attached to the camera. This combo will be placed across the width of the compartment (from side to side of the bag). The prime lens will sit on its circumference and might fit along side of the zoom lens (pending the offset of the zoom on the camera body). A partition has to be used to stop the two lenses from contact. The fit would be very close and I would recommend trying this bag out to make sure that the zipper will close and the fit is acceptable to your personal preference.

Is it padded enough for caring camera during Mountain and Road Bike riding? Would gear survive in case of crash?

I have not worn the pack for cycling, but doubt it would protect the gear in a crash. You might search for a specialty pack for that level of protection. It is a nice comfortable daypack for the city, light day hiking and travel in my opinion.

We put an IPad with it's case in the provided padded pouch in this pack with no problems. It measures 9 1/2 X 7 1/2 X 3/4 .I have no idea what size you laptop is, but the provided space will not fit anything much bigger.
