It depends on the output connector of the audio interface, and possibly to a lesser extent the input connectors on the headphone amp. If the audio interface's connector is a stereo output, it might work; it would depend on whether the headphone amp can accept unbalanced connections (my guess is most amps should accept it). But if the audio interface's connector is a single channel (probably capable of unbalanced or balanced connections), then you'll only get a sound signal on the tip of the stereo hosa connector, and only on whatever channel you connect the corresponding mono connector to the headphone amp.
