I contacted Sabrent customer support about this question. Here's their reply:
"Let me mention that it should operate at PCIe 4.0 x4 speeds, but it depends on how the slots are traced/chipset or how the lanes are distributed, motherboards may have issues at times allocating those lanes, please check if that bottom slot is tied to an M.2 slot sometimes, so that could also result in slower speeds or worse, an unstable connection or no detection we recommend using the appropriate slot of your motherboard but it is something that you check with the manufacturer of your motherboard. "
