Is this lens build strong enough to support the weight of a 100mm filter system such as the Nisi 100mm filter system?

The Canon RF 16mm f/2.8 STM Lens uses 43mm front filter threads. Unfortunately, none of the 100mm (4-inch) filter holders we carry have 43mm adapter rings to connect the filter holder to the lens' filter threads. While it may be possible to use a step-up ring to connect the filter holder to the lens, I would probably try to use a different lens.

It is not written on the lens but my internal system says the country of origin is Taiwan.

Is there both auto focus and manual focus on this lens? I do not see a focus circle for manual focus on the lens.

This lens does have AF and MF as options. The switch on the lens can be used to control focus or to use as a custom configuration. When it's set to Focus, and you set the camera's focusing mode to MF, the ring will control the focus.

When I added 52 mm ND filters, I encountered horrendous vignetting as I got to the stronger densities. I am here looking for solutions to this. Maybe go even larger in thread size?

Is lens will correctly set with dji rsc2 plate? Plate does not fall into the frame?

16mm on full frame is pretty wide. I think you will be okay because with how much the camera protrudes from the rear mount portion of the plate and the front optics, it looks like it should be okay not getting the bottom of the RSC2 plate in the frame. Note, it has not been tested. But if it, does it will be a small amount and you can crop it.
