Have anyone tried this RF extender on a RF body, then an EF adapter, finally, an EF lense?
In theory, it should work...
No, they are not compatible. All Canon RF to EF adapters have a narrow square shape opening on the back; therefore, the Canon Extender RF 2x front optic cannot fit through the adapter.

Can you safely use a 2x teleconverter with extension tubes(all Canon original)?
Just curious!
Extension tubes will fit the 2x converter but how well they work would be unknown. The tube)s) would need to be mounted to the camera, then the 2x converter, and then the compatible lens. Automatic functions such as AF and auto exposure might be lost too.

If used with 600m lens it stops to f22, does that ...
If used with 600m lens it stops to f22, does that effect the camera settings e.g. shutter speed and ISO?
Yes and no. To maintain a correct exposure, your exposure settings will have to compensate for the loss of light. With the 2.0x teleconverter, you lose 2-stops of light.
For the "no" answer: in manual, since you have to change these settings yourself, if you do not change your shutter speed or ISO to compensate, your picture will come out 2-stops darker. So if you decide not to change them, technically the lens and teleconverter do not have any control over your other settings.
For the "yes" answer: in P/Tv/Av, your shutter speed and ISO will adjust accordingly to your EV compensation, min. shutter speed settings, and also Auto ISO ceiling (if applicable).
