You can plug your audio mixer's main output straight into the S1 Pro+, yes. The best input to use for this would actually be the small headphone port (between "AUX" and the Bluetooth button). This will keep Input 1 and Input 2 free.

What are some compatible Wireless Headset Mics that will work with the XLR Transmitter?

Any dynamic headset microphone, such as the Shure WH20 Headset Mic (B&H # SHWH20XLR) or the Audio-Technica PRO 8-HEX Dynamic Headworn Mic (B&H # AUPRO8HEX), is compatible for use with the XLR transmitter included with the Bose S1 Pro+ Wireless PA System Kit with Mic/Line and Instrument Transmitters. Since the Bose transmitter does not supply phantom power or plug-in power, it will not work with most condenser microphones, unless the mic has its own power source, such as a battery, to supply the voltage that a condenser microphone requires.

Are there any issues with pairing this version of the Bose S1 Pro + with the original version?

As the S1 Pro system uses the Bose Connect app and the S1 Pro+ system uses the Bose app, it's not possible to connect these two systems wirelessly. However, a cabled solution does exist; it would be possible to take a LINE OUT from one speaker and run this into the 3.5mm stereo input on the other speaker. This would keep Channels 1 and 2 completely free on the second speaker.

Can Bose S1 Pro+ interface with computer for Zoom or video transmission. What add'l equipment is needed?

When using the Bose S1 Pro+ Wireless PA System Kit with Sennheiser Handheld Mic and Mic Transmitter for this application, the wireless mic system would only work with the speaker directly. It will not work to transmit that audio to the other side of the Zoom call. Furthermore, you would need to use an audio interface to output the audio from the Zoom call to the Bose S1 Pro+ speaker, and to be able to connect an external microphone so that the microphone can be heard on the other side of the zoom call and the Bose S1 Pro+.

The S1 Pro+ can be positioned in an angled up right or angled side position. Optionally it can be mounted to a 1.38" post on a tripod via the integrated pole cup.
